MSlesionsarecharacterizedbydestructionofmyelinandsignificantneuronalandaxonalloss.PreliminarystudieswiththeuseofTregsinthemousemodelofMShavebeenextremelyencouraging.However,recentstudieswithhumancellshaveshownthepresenceofdifferentsubpopulationsofTcellswithintheCD4+CD25+Foxp3+Tcellphenotype,someofwhichdonothaveregulatoryfunctions.Thesefindingssuggestapotentialdifferencebetweenmouseandhumanintheregulatoryphenotype.Here,weshowthathumanactivatedCD4+CD25+Foxp3+Tcellsareneurotoxicinvitro.ThesecellsexpressedhighlevelsofthecytotoxicmoleculeGrBandhadnosuppressiveeffect.Onthecontrary,theyproducedIFN‐γandlowIL‐17,suggestingashifttowardaTH1phenotype.Thus,ourdataconfirmthepresenceofanonregulatorycytotoxicsubpopulationwithinthehumanCD4+CD25+Foxp3+Tcellsandsuggestfurtherstudiesonthehumanregulatoryphenotypepriortoanypotentialtherapeuticapplication...
