M1/M2cytokine‐dependentpolarizationofprimaryhumanMDMshasbeenshowntocontainCCR5‐dependent(R5)HIV‐1replication.Inthisstudy,asimilareffectwasachievedwhenmonocyteswerefirstpolarizedtowardM1orM2andwereinfected7daftertheirdifferentiationintoMDMs,regardlessofwhetherthecytokineswereremoved18haftercellstimulationorwereleftinculture.UnlikepolarizedMDMs,nosignificantdown‐regulationofCD4fromthecellsurfacewasobservedinMDMsderivedfromM1/M2‐polarizedmonocytes.AsecondstimulationofMDMsdifferentiatedfromM1/M2monocyteswiththeoppositepolarizingcytokinesconvertedthevirusreplicationprofileaccordingtothenewstimuli.TheexpressionofM1andM2markers(i.e.,APOBEC3AandDC‐SIGN,respectively)wasinducedbyMDMstimulationwiththeoppositecytokines,althoughitalsopersistedincellsaccordingtotheirfirststimulatorycondition.Thus,stimulationofmonocyteswithM1‐andM2‐inducingcytokinesleadstoarestrictionofHIV‐1replicationwhenthesecellsareinfectedseveraldayslaterasdifferentiatedMDMs.TheseobservationsimplythatactivationofcirculatingmonocytessignificantlyinfluencestheircapacitytoeithersupportorrestrictHIV‐1replication,onceextravasated,andeventuallytobecomeinfectedastissuemacrophages...
