Inflammationaltershematopoiesis,oftenbydecreasingerythropoiesisandenhancingmyeloidoutput.ThemechanismsbehindthesechangesandhowtheBMstromacontributestothisprocessareactiveareasofresearch.Inthisstudy,weexaminethesequestionsinthesettingofmurineToxoplasmagondiiinfection.Ourdatarevealthatinfectionaltersearlymyeloerythroiddifferentiation,blockingerythroiddevelopmentbeyondthePreMegEstage,whileexpandingtheGMPpopulation.IL‐6wasfoundtobeacriticalmediatorofthesedifferences,independentofhepcidin‐inducedironrestriction.ComparingtheBMwiththespleenshowedthatthehematopoieticresponsewasdrivenbythelocalmicroenvironment,andBMchimerasdemonstratedthatradioresistantcellsweretherelevantsourceofIL‐6invivo.Finally,directexvivosortingrevealedthatVCAM+CD146loBMstromalfibroblastssignificantlyincreaseIL‐6secretionafterinfection.ThesedatasuggestthatBMSCsregulatethehematopoieticchangesduringinflammationviaIL‐6...
