Wereviewtheemergingconceptthatchangesincellularbioenergeticsconcomitantlyreprograminflammatoryandmetabolicresponses.Themolecularpathwaysofthisintegrativeprocessmodifyinnateandadaptiveimmunereactionsassociatedwithinflammation,aswellasinfluencingthephysiologyofadjacenttissueandorgans.Theinitiatingproinflammatoryphaseofinflammationisanabolicandrequiresglucoseastheprimaryfuel,whereastheopposingadaptationphaseiscatabolicandrequiresfattyacidoxidation.ThefuelswitchtofattyacidoxidationdependsonthesensingofAMPandNAD+byAMPKandtheSirTfamilyofdeacetylases(e.g.,SirT1,‐6,and‐3),respectively,whichcoupleinflammationandmetabolismbychromatinandproteinreprogramming.TheAMP‐AMPK/NAD+‐SirTaxisproceedssequentiallyduringacutesystemicinflammationassociatedwithsepsisbutceasesduringchronicinflammationassociatedwithdiabetes,obesity,andatherosclerosis.Rebalancingbioenergeticsresolvesinflammation.Manipulatingcellularbioenergeticsisidentifyingnewwaystotreatinflammatoryandimmunediseases...
