TheproinflammatoryactivitiesofIL‐1aretightlycontrolledatdifferentlevels.IL‐1R2actsasadecoyreceptorandhasbeenshowntoregulatethebiologicaleffectsofIL‐1invitroandinvivo.However,littleisknownaboutitsnaturalexpressioninthemouseinphysiologicandpathologicconditions.Inthisstudy,weexaminedIL‐1R2mRNAandproteinexpressioninisolatedcellsandtissuesinresponsetodifferentstimulatoryconditions.DataobtainedusingexvivoCD11b+Ly6G+peripheralbloodcellsandinvitro‐differentiatedCD11b+Ly6G+BMGindicatedthatneutrophilsarethemajorsourceofconstitutivelyexpressedIL‐1R2inthemouse.TheexpressionofIL‐1R2onBMGandexvivoLy6G+peripheralbloodcellswashighlyup‐regulatedbyHC.IL‐1R2pull‐downexperimentsshowedthatmouserIL‐1βbindstoBMGIL‐1R2,whereasbindingofIL‐1Racouldnotbedetected.Furthermore,LPStreatmentinducedsheddingofIL‐1R2fromtheneutrophilmembraneinvitroandinvivo,executedmainlybyADAM17.Finally,ininvivomodelsofinflammation,includingthioglycolate‐inducedacuteperitonitisandacutelunginjury,infiltratingLy6G+neutrophils,expressedIL‐1R2.Ourdatashowthatinthemouse,neutrophilsmainlyexpressthedecoyreceptorIL‐1R2undernaïveandinflammatoryconditions.Thesedatasuggestthatneutrophilsmaycontributetotheresolutionofacuteinflammation...
