Thisreviewsummarizesthecurrentstateofknowledgeregardingtherolesofthesignalingadapterproteintumornecrosisfactorreceptor(TNFR)‐associatedfactor3inregulatingthefunctionsofBandTlymphocytes.InBlymphocytes,TNFR‐associatedfactor3inhibitssignalingbyTNFRsuperfamilyreceptors,Toll‐likereceptors,andinterleukin‐6R.Incontrast,signalingtoBcellsbythevirallyencodedoncogenicproteinlatentmembraneprotein1ispromotedbyTNFR‐associatedfactor3.AnimportantBcell‐specificroleforTNFR‐associatedfactor3istheinhibitionofhomeostaticsurvival,directlyrelevanttothecommonoccurrenceofTNFR‐associatedfactor3mutationsinhumanBcellmalignancies.TNFR‐associatedfactor3wasrecentlyfoundtobearesidentnuclearproteininBcells,whereitinteractswithandinhibitsgeneexpressionmediatedbythecAMPresponseelement‐bindingproteintranscriptioncomplex,includingexpressionoftheprosurvivalproteinmyeloidleukemiacelldifferentiationprotein1.InTlymphocytes,TNFR‐associatedfactor3isrequiredfornormalsignalingbytheTcellantigenreceptor,whileinhibitingsignalingbytheinterleukin‐2receptor.CytoplasmicTNFR‐associatedfactor3restrainsnuclearfactor‐κB2activationinbothTandBcells.Clinicalimplicationsandfuturedirectionsforthestudyofthiscontext‐dependentsignalingregulatorarediscussed...
