【 摘 要 】
Nano‐sizedmembranevesiclesaresecretedbymanycelltypes.Thesevesiclescanserveascarriersofcellularinformation.DC‐derivedvesiclescanbetargetedtootherimmunecellsandmodifytheirfunction.AccurateanalysisofquantitativeandqualitativechangesinEVproductionbyDCupondifferentactivationstimuliisneededtofurtherrevealtheimmuneregulatorypropertiesofDC‐derivedEVs.However,methodsforreliablequantificationofindividualEVsandforanalysisoftheheterogeneityofEVpopulationsarelimited.Withourrecentlydevelopedhigh‐resolutionflowcytometry‐basedmethod,wecanperformahigh‐throughput,multiparameter,andquantitativeanalysisofindividualEVs.Withtheuseofthisnoveltechnique,weshowthatdespitepreviousassumptions,stimulationwithbacterialLPSincreasesEVreleasebyDC.Furthermore,wedemonstrateheterogeneityinDC‐derivedEVsregardingtheirbuoyantdensityandMHCclassIIcontent.Finally,weshowthatcognateinteractionbetweenLPS‐stimulatedDCandCD4+TcellsaffectsboththequantityandqualityofLPSDC‐derivedEVspresentintheculturesupernatant.Thesedataindicatethatflowcytometry‐basedanalysisofindividualEVsisavaluable,noveltooltostudythedynamicsofEVsecretionandcomposition,offeringgreatopportunitiestounveilthefunctionofimmunecell‐derivedEVs...
