【 摘 要 】

Apreviouslyunsuspected,considerableproportionofnewlysynthesizedpolypeptidesarehydrolyzedrapidlybyproteasomes,possiblycompetingwithendogenoussubstratesandalteringproteostasis.Inviewoftheanti‐cancereffectsofPIs,wesetouttoachieveaquantitativeassessmentofproteasomeworkloadincellshallmarkedbydifferentPIsensitivity,namely,apanelofMMcells,andinadynamicmodelofplasmacelldifferentiation,aprocessthatconfersexquisitePIsensitivity.OurresultssuggestthatproteinsynthesisisakeydeterminantofproteasomalproteolyticburdenandPIsensitivity.IndifferentMMcellsandindifferentiatingplasmacells,theaverageproteolyticworkaccomplishedperproteasomerangesoverdifferentordersofmagnitude,anunexpecteddegreeofvariability,withincreasedworkloadinvariablyassociatedtoincreasedPIsensitivity.Theunfavorableload‐versus‐capacitybalancefoundinhighlyPI‐sensitiveMMlinesisaccountedforbyadecreasedtotalnumberofimmunoproteasomes/cellcoupledtoenhancedgenerationofRDPs.Moreover,indicativeofcause‐effectrelationships,attenuatinggeneralproteinsynthesisbytheotherwisetoxicagentCHXreducesPIsensitivityinactivatedBandinMMcells.OurdatasupporttheviewthatinplasmacellsproteinsynthesiscontributestodeterminePIsensitivitybysaturatingtheproteasomaldegradativecapacity.Quantitatingproteinsynthesisandproteasomeworkloadmaythusprovecrucialtodesignnovelnegativeproteostasisregulatorsagainstcancer...
