EEisanemergingdiseasereportedinchildrenandadultsofurbanizedcountries,whereindoorinsectallergensaremajorhealthriskfactors.ReviewofourhospitalpatientdatabaseuncoveredthatanumberofEEpatientshavehypersensitivitytoindoorcat,dog,cockroach,anddustmiteallergens.WetestedthehypothesiswhetherinhaledindoorinsectallergensareeffectiveinducersofexperimentalEE.Wedeliveredcat,dog,cockroach,anddustmiteallergenextractsintranasallytowild‐typeandeotaxin‐1/2‐,CCR3‐,andIL‐5‐deficientmice.Interestingly,wild‐typemiceexposedtocockroachordustmiteallergensdevelopasignificantincreaseinthelevelsofesophagealeosinophilsandmastcellscomparedwithsaline‐challengedmice.Theeosinophilnumbersintheesophagusofcockroach‐anddustmite‐exposedmicewere18.3±6.8/mm2and33.4±11.1/mm2comparedwith2.3±1.8/mm2and2.1±1.2/mm2insaline‐challengedmice.Additionally,weobservedanadditiveeffectofthesetwoallergensininducingesophagealeosinophiliaandmastocytosis.Histopathologicalanalysisdetectedintraepithelialesophagealeosinophiliainmiceexposedtobothallergens.Furthermore,miceexposedtocockroachand/ordustmitehadincreasedlevelsoftotalIgEandantigen‐specificIgG1inthebloodandincreasedesophagealexpressionofeosinophil‐activecytokines(IL‐13)andchemokines(eotaxin‐1).Notably,micedeficientineotaxin‐1/2,CCR3,andIL‐5showedablatedesophagealeosinophiliafollowingcockroachordustmiteallergenexposure.ThesedataindicatethatindoorinsectallergensarepotentinducersofIL‐5andeotaxin‐mediatedesophagealeosinophilia.Theseexperimentalstudiesareinaccordancewithclinicaldatabutmayhavesomelimitationsinherenttoanimalmodelsofhumandisease...
