ColonizationbySAisassociatedwithexacerbationofAD.EosinophilicinflammationisacardinalpathologicalfeatureofAD,butlittleisknownaboutpossibledirectinteractionbetweenSAandeosinophils.PAFRappearstobeinvolvedinphagocytosisofGram‐positivebacteriabyleukocytes.TheobjectiveofthisstudywastoinvestigatewhetherSAdirectlyinduceseosinophileffectorfunctionsviaPAFRinthecontextofADpathogenesis.Peripheralbloodeosinophilswereculturedwithheat‐killedSA,andEDNrelease,superoxidegeneration,andadhesiontofibronectin‐coatedplatesweremeasured.Cytokines,releasedinthesupernatants,werequantifiedbymultiplexbeadimmunoassays.FISH‐labeledSAwasincubatedwitheosinophilsandvisualizedbyconfocallaser‐scanningmicroscopy.PAFR‐blockingpeptideandPAFRantagonistsweretestedforinhibitoryeffectsonSA‐inducedreactions.SAinducedEDNreleaseandsuperoxidegenerationbyeosinophilsinadose‐dependentmanner.IL‐5significantlyenhancedSA‐inducedEDNrelease.IL‐5andIL‐17AsignificantlyenhancedSA‐inducedsuperoxidegeneration.SAenhancedeosinophiladhesiontofibronectin,whichwasblockedbyanti‐CD49d,andinducedeosinophilsecretionofvariouscytokines/chemokines(IL‐2R,IL‐9,TNFR,IL‐1β,IL‐17A,IP‐10,TNF‐α,PDGF‐bb,VEGF,andFGF‐basic).AfterincubationofeosinophilswithSA,FISH‐labeledSAwasvisualizedintheeosinophilsˈcytoplasm,indicatingphagocytosis.APAFR‐blockingpeptideandtwoPAFRantagonistscompletelyinhibitedthosereactions.Inconclusion,SAdirectlyinducedeosinophilactivationviaPAFR.BlockadeofPAFRmaybeanovel,therapeuticapproachforADcolonizedbySA...
