【 摘 要 】
TheendothelialdamageinducedbyStxrepresentsthemainpathogeniceventintheHUSassociatedwithSTECinfectionsinhumans.Stx,releasedinthegutbybacteria,enterthebloodstreamandaretargetedtorenalendothelia.TheroleofPMNasatoxincarrierhasbeentheobjectofcontroversy.Inthispaper,weconfirmthebindingofStx1toPMN,alsoshowingitsdegranulatingeffectsonfull‐loadedleukocytes,andsupportthecarrierroleofPMNbyusingatwo‐chambertransmigrationdevice,inwhichPMN,loadedinvitrowithdifferentamountsofStx1,transmigratedthroughconfluentmonolayersofendothelialcells,mimickingthetoxin‐inducedrenalendothelialinjury.Stx1wastransferredduringPMNtransmigration,impairingproteinsynthesisandtriggeringproductionofproinflammatorycytokinesinendothelialcells.PMN,carryinglowtoxinamounts,inducedthereleaseofhighlevelsofcytokinesinviableendothelialcells,whereascytokineproductionwasblockedincellschallengedwithPMNfullyloadedwithStxasaresultofanalmosttotalimpairmentoftranslationandoftheactivationoftheapoptoticprogram.Inagreementwithpreviousunexplainedobservationsinanimalmodels,theresultsobtainedwithourexperimentalsettingsuggestthataself‐amplifyingcircletriggeredbylowdosesoftoxinmayleadtotheproductionofproinflammatorymediatorsofrenaldamageinHUS...
