Differential accumulation and function of proinflammatory 6‐sulfo LacNAc dendritic cells in lymph node and colon of Crohn's versus ulcerative colitis patients

【 摘 要 】

HumanSlanDCshavebeenstudiedinpatientswithpsoriasis,rheumatoidarthritis,cancer,andautoimmunediseases.Inthisstudy,weinvestigatedthefrequency,phenotype,andfunctionofSlanDCsinblood,colon,aswellasmLNsofpatientswithIBD.WefirstshowthatthefrequencyofcirculatingCD14dullSlanDCswasreducedinCDpatientsrefractorytoimmunosuppressivedrugsorTNF‐αblockersrelativetountreatedCD,UC,andhealthysubjects.InbloodofCDpatients,SlanDCsexpressedCD172a,asdetectedbyCD47fusionproteinbinding,whencomparedwithitslackofexpressionincontrolsubjects.Next,wedemonstratethatCD172a+SlanDCsthatproducedIL‐1βandTNF‐αaccumulatedinmLNsandcolonsofCDpatients.TheCD172a+SlanDCsup‐regulatedtheirexpressionofCD14inCDtissuesandtheproinflammatorycytokineswereproducedinCD14brightCD172a+SlanDCs.Bycontrast,nodifferencewasnotedinthefrequencyofSlanDCsbetweeninflamed,noninflamedcolonicmucosaofUCpatientsandcontrol,non‐IBDdonors.Finally,thepercentageofcytokine‐producingSlanDCsalsoaugmentedinresponsetoTLR2andNOD2ininvitrostimulationinPBMCsofCD,butnotUC,patients.Inconclusion,weproposethatproinflammatoryCD14brightCD172a+SlanDCsareadistinguishingfeaturebetweenCDandUC,asthesecellsaccumulateuniquelyinmLNsandcolonicmucosaofCDpatients.Thus,SlanDCsmaycontributetoCDimmunopathogenesis...
