ThedynamicsofimmunecellpopulationsduringacuteHIV‐1infectionarenotfullydeciphered,especiallyfornon‐Tcells.Inthisstudy,wetestedwhetherspecificcellularsubsetsoftheinnatearmoftheimmuneresponseareaffectedearlyafterHIV‐1infection.UsingacohortofHIV‐1‐infectedindividuals,wehavemonitoredtherelativefrequencyofbloodTlymphocytes,monocytes,andDCsatvariousinfectionstagesandmeasuredtheirrespectiveintracellularHIV‐1DNAloads.TheHIV‐1DNAloadinnaiveCD4+Tlymphocytes,whicharelostveryearlyduringacuteinfection,wasten‐to100‐foldlowerthaninCD57–andCD57+memoryCD4+Tlymphocytes.Weobservedthatdespiterapid,persistentlossafterHIV‐1infection,pDCsrepresentedanon‐negligibleHIV‐1DNAreservoir.CD16+proinflammatorycDCsandmonocytesaccumulatedgradually,andHIV‐infectedCD16+monocytescontainedhigherHIV‐1DNAloadsthantheirCD16–counterpartduringacuteinfection.Duringchronicinfection,CD16+cDCsexhibitedhigherHIV‐1DNAloadsthantheCD16–population.Overall,ourresultsdemonstratethatnon‐TcellcompartmentsareamajorHIV‐1DNAreservoir,andCD16+monocytesandCD16+cDCspotentiallyplayanimportantroleinHIV‐1dissemination...
