HO‐1istheonlyinducibleoneofthreeisoenzymesthatcatalyzestheoxidativedegradationofheme.HO‐1isinduciblebyvariouscellularstressfactorsandexertscytoprotectiveandimmunomodulatoryeffects.RecentpublicationsdemonstratedthatHO‐1isconstitutivelyexpressedbyCD4+CD25+TregsandinducedinCD4+CD25−TcellsuponFoxP3transfection.Here,weinvestigatedwhetherHO‐1wasessentialandsufficientforhumanTregstoexertimmunosuppressioninvitro.PGJ2inducedpronouncedexpressionofHO‐1inCD4+CD25−TcellswithoutaccompanyingFoxP3induction.TreatmentofCD4+CD25−TcellswithPGJ2decreasedtheirproliferation,whereastheHO‐1inhibitorSnPPenhancedtheproliferationofHO‐1‐expressingTregs,suggestingthatHO‐1maymodulatetheproliferativecapacityofTlymphocytes.HO‐1modulationbySnPPtreatmentofTregsorPGJ2treatmentofCD4+CD25−Tcellsneithersuppressednorinducedimmune‐modulatoryfunctioninthesecells,respectively,asmeasuredbyresponder‐cellproliferationand/orIL‐2production.Insummary,thesedatasuggestthatHO‐1expressionbyTregsmightcontributetotheirtypicalreluctancetoproliferatebutdoesnotaccountindependentlyfortheirsuppressivefunctions...
