Immunosuppressioniscurrentlythetreatmentofchoicetoattenuatethechronicdeteriorationoftissuefunctionasaresultoftheeffectormechanismsoftheimmunologicalresponseintransplantrejectionandautoimmunediseases.However,globalimmunosuppressiongreatlyincreasestheriskofacquiringlife‐threateninginfectionsandisassociatedwithorgantoxicitywhenusedlong‐term.Thus,alternativeapproachesthatinhibitonlytheunwantedimmuneresponsesandpreservegeneralimmunityarehighlydesirable.Thereceptor/ligandpairsinvolvedinthecross‐talkbetweenDCandTcellshavebeenthefocusofintenseandexcitingresearchduringthelastdecade.TheHVEM/LIGHT/BTLA/CD160costimulatory/coinhibitorypathwayhasemergedasapotentialtargetforthedevelopmentofimmunetherapeuticinterventions.Herein,wewillsummarizeanddiscusshowblockadeofthecostimulatoryHVEM/LIGHTinteractionoragonistsignalingthroughtheinhibitoryBTLAandCD160receptorscouldcontributetothecontrolofdeleteriousimmuneresponses...
