PsoriasisisaninflammatorycutaneousdisordercharacterizedbymarkedepidermalthickeningandTh1andTh17cellinfiltration.Atpresent,thecontributionofBcellstothepathogenesisofpsoriasisisunclear.Inmice,topicalapplicationofimiquimodinducesinflamedskinlesionsandservesasanexperimentalanimalmodelforhumanpsoriasis.Inthisstudy,weshowedthatimiquimod‐inducedskininflammationwasmoresevereinCD19−/−thanWTmice.TheseinflammatoryresponseswerenegativelyregulatedbyauniqueIL‐10‐producingCD1dhiCD5+regulatoryBcellsubset(B10cells)thatwasabsentinCD19−/−miceandrepresentedonly1–2%ofsplenicB220+cellsinWTmice.SplenicB10cellsenteredthecirculationandmigratedtodrainingLNsduringimiquimod‐inducedskininflammation,therebysuppressingIFN‐γandIL‐17production.Furthermore,adoptivetransferoftheseB10cellsfromWTmicereducedinflammationinCD19−/−mice.ThepresentfindingsprovidedirectevidencethatB10cellsregulateimiquimod‐inducedskininflammationandofferinsightsintoregulatoryBcell‐basedtherapiesforthetreatmentofpsoriasis...
