Macrophagesarenowroutinelycategorizedintophenotypicsubtypesbasedongeneexpressioninducedinresponsetocytokineandpathogen‐derivedstimulation.Inthebroadestdivision,macrophagesaredescribedasbeingCAMs(M1macrophages)orAAMs(M2macrophages)basedontheirexposuretoTLRandIFNsignalsorTh2cytokines,respectively.Despitetheprolificuseofthissimpleclassificationscheme,littleisknownabouttheprecisefunctionsofeffectormoleculesproducedbyAAMs,especiallyhowrepresentativetheCAMandAAMsubtypesareoftissuemacrophagesinhomeostasis,infection,ortissuerepairandhowplasticityingeneexpressionregulatesmacrophagefunctioninvivo.Furthermore,correlationsbetweenmouseandhumantissuemacrophagesandtheirrepresentativesubtypesarelackingandareamajorbarriertounderstandinghumanimmunity.Here,webrieflysummarizecurrentfeaturesofmacrophagepolarizationanddiscusstherolesofvariousmacrophagesubpopulationsandmacrophage‐associatedgenesinhealthanddisease...
