TcellsproduceanumberofcytokinesandchemokinesuponstimulationwithTLRagonistsinthepresenceorabsenceofTCRsignals.Here,weshowthatsecretionofneutrophilchemoattractantCXCL8fromhumanTcelllineJurkatinresponsetostimulationwithTLRagonistsisreducedwhencellstimulationiscarriedoutinpresenceofserum.Serumdoesnot,however,inhibitTCR‐activatedsecretionofCXCL8nordoesitdown‐regulateTLR‐costimulatedIL‐2secretionfromactivatedTcells.ThemoleculethatcanmimictheabilitytobringaboutsuppressioninCXCL8fromTLR‐activatedTcellsisserum‐bornebioactivelipid,S1P.SerumandS1P‐mediatedinhibitionrequireintracellularcalcium.S1PalsosuppressesCXCL8secretionfromperipheralblood‐derivedhumanTcellsactivatedexvivowithvariousTLRligands.OurfindingsrevealapreviouslyunrecognizedroleforS1PinregulatingTLR‐inducedCXCL8secretionfromhumanTcells...
