【 摘 要 】
IL‐10isakeyimmune‐regulatorycytokine,anditsgenepolymorphismscorrelatewithseverityofclinicalGVHD.IL‐10ismadebyavarietyofdonorandhostcells,butthefunctionalrelevanceofitssourceanditsroleinthebiologyofacuteGVHDarenotwellunderstood.WeusedpreclinicalmodelstoexaminetherelevanceofIL‐10−/−indonorandhostcellularsubsetsontheseverityofGVHD.IL‐10−/−inhosttissuesorinthedonorgraftsdidnotalterdonorTeff‐mediatedseverityofGVHD.Furthermore,neitherhost‐derivednordonorTeff‐derivedIL‐10wasrequiredforregulationofGVHDbyWTCD4+CD25+donorTregs.Bycontrast,Treg‐derivedIL‐10,althoughnotobligatory,wasnecessaryforoptimalreductionofGVHDbymaturedonorTregs.Importantly,IL‐10fromdonorBMgraftswasalsocriticalforoptimaldonorTreg‐mediatedsuppressionofGVHD.Together,thesedatasuggestthatIL‐10doesnotcontributetotheinductionofGVHDseveritybytheTeffs.However,donorBMgraftandTreg‐derivedIL‐10areimportantfordonorTreg‐mediatedsuppressionofGVHD...
