Eosinophilsaregranulocytesassociatedwithhostdefenseagainstparasitichelminthswithallergicconditionsandmorerecently,withimmunoregulatoryresponses.Eosinophilsaredistinguishedfromleukocytesbytheirdominantpopulationofcytoplasmiccrystalloid(alsotermedsecretory,specific,orsecondary)granulesthatcontainrobuststoresofdiverse,preformedcationicproteins.Here,weprovideanupdateonourknowledgeabouttheuniqueandcomplexstructureofhumaneosinophilcrystalloidgranules.Wediscusstheirsignificanceasrichsitesofavarietyofreceptorsandreviewourownrecentresearchfindingsandthoseofothersthathighlightdiscoveriesconcerningthefunctionofintracellularreceptorsandtheirpotentialimplicationsincellsignaling.Specialfocusisprovidedonhoweosinophilsmightusetheseintracellularreceptorsasmechanismstosecrete,selectivelyandrapidly,cytokinesorchemokinesandenablecell‐freeextracellulareosinophilgranulestofunctionasindependentsecretorystructures.Potentialrolesofcell‐freeeosinophilgranulesasimmuneplayersintheabsenceofintacteosinophilswillalsobediscussed...
