pDCarethemostpotentIFN‐α‐producingcellsinthebodyandserveasavitallinkbetweeninnateandadaptiveimmunity.DeficienciesinpDCfunctionwereamongtheearliestobservationsofimmunedysfunctioninHIV‐1infection.Herein,wereviewthestatusofpDCinindividualswithHIV‐1infectionandthepotentialroleofthesecellsinpathogenesis.WebeginbyreviewingthebasicpropertiesofpDCandthendiscussthecompromiseincirculatingpDCnumbersandfunctioninearlyandviremicHIV‐1infectionandmechanismsthatmightaccountfortheirdepletioninHIV‐infectedpatients.Inaddition,wereviewtheevidencethatchronicproductionofIFN‐α,probablythroughthechronicactivationofpDC,iscentraltotheimmuneactivationthatissodetrimentalinHIVinfection.Finally,wediscusstheimportanceofbalanceinpDCnumbersandfunctionandthepotentialvalueofusingabsolutepDCcountsandfunctionasabiomarker,alongwithCD4+cellcountsandVLinHIV‐1‐infectedpatients...
