【 摘 要 】
Duringevolution,theimmunesystemhasdiversifiedtoprotectthehostfromtheextremelywidearrayofpossiblepathogens.Untilrecently,immuneresponsesweredissectedbyuseofglobalapproachesandbulktools,averagingresponsesacrosssamplesandpotentiallymissingparticularcontributionsofindividualcells.Thisisastronglylimitingfactor,consideringthatinitialimmuneresponsesarelikelytobetriggeredbyarestrictednumberofcellsatthevanguardofhostdefenses.Thedevelopmentofnovel,single‐celltechnologiesisamajorinnovationofferinggreatpromiseforbasicandtranslationalimmunologywiththepotentialtoovercomesomeofthelimitationsoftraditionalresearchtools,suchaspolychromaticflowcytometryormicroscopy‐basedmethods.Atthetranscriptionallevel,muchprogresshasbeenmadeinthefieldsofmicrofluidicsandsingle‐cellRNAsequencing.Attheproteinlevel,masscytometryalreadyallowstheanalysisoftwiceasmanyparametersasflowcytometry.Inthisreview,weexplorethebasisandoutcomeofimmune‐celldiversity,howgeneticallyidenticalcellsbecomefunctionallydifferent,andtheconsequencesfortheexplorationofhost‐immunedefenseresponses.Wewillhighlighttheadvantages,trade‐offs,andpotentialpitfallsofemerging,single‐cell‐basedtechnologiesandhowtheyprovideunprecedenteddetailofimmuneresponses...
