GCsarepowerfulanti‐inflammatorycompoundsinhibitinginflammatorycellrecruitmentandproductionofproinflammatorycytokines.WehaverecentlyfoundthatDCs,thekeyplayersofTcellprimingandpolarization,respondtoallogeneicapoptoticneutrophilswithproinflammatorycytokinereleaseandTh1cellactivation.Here,weshowthatmonocyte‐derivedhumanDCsdeveloptheircapacitytoengulfapoptoticcellsbyup‐regulatingasetofapoptophagocyticgenes.ThisgeneexpressionpatternwasreprogrammedwhendifferentiationtookplaceinthepresenceofthesyntheticGCDex,whichincreasedtheexpressionofphagocytosisreceptorsMERTKandCD14,thebridgingmoleculeC1QA,DNASE2,andADORA3.TheincreasedphagocytosiswasattenuatedbytheadditionofADORA3antagonistandcouldnotbeobservedwhenbonemarrow‐derivedDCsofADORA3KOmiceweretreatedwithDex.TheGC‐treatedhumanDCsloadedwithallogeneicapoptoticneutrophilssecreted,inresponsetoLPSandIFN‐γ,theinflammatorycytokineTNF‐α.Furthermore,theDex‐treatedDCscouldactivateautologousTlymphocytestowardTh1effectorcells,andthiswasenhancedbytheirexposuretoallogeneicapoptoticneutrophils...
