Inflammationisthebody'sinitialresponsetoinfection,whichisharmfulwhenexcessive,asexemplifiedinsepsisinflammatorysyndromes.EctodomainsheddingbythemembranemetalloproteaseADAM17isanemergingregulatorofinflammation,asitdirectstheactivityofvariousinflammatorymodulators.Atthistime,however,littleisknownabouttheinvivofunctionofADAM17.Here,weshowthatADAM17‐deficientleukocytesaffordedmiceasurvivalbenefitfollowingEscherichiacoli‐mediatedperitonealsepsis,whichwasassociatedwithareductioninsystemicproinflammatorycytokinelevelsandbacterialburden.AmorerapidyettransitoryneutrophilinfiltrationintotheperitonealcavityofconditionalADAM17knockoutmicewasobservedwhencomparedwithcontrolmice,suggestingamechanismfortheirenhancedclearanceofbacteria.PreventingthesheddingofL‐selectinaugmentsneutrophilrecruitment,andweshowthatL‐selectinsheddingbyperitonealneutrophilsinconditionalADAM17knockoutmicewasimpaired.Moreover,theirperitonealTNF‐αlevelsweremarkedlylowerthancontrolmicefollowingE.colichallenge.TheseeventsindicatekeymolecularprocessesinvolvedinthealteredtimecourseofneutrophilrecruitmentinconditionalADAM17knockoutmice.Overall,ourstudyprovidesnovelinvivoevidenceoftheinstrumentalroleofADAM17inmodulatinginflammationandhostresistanceduringGram‐negativebacterialinfection...
