EVsinthemicrovesiclesizerangereleasedduringspontaneousdeathofhumanneutrophilswerecharacterizedandtheirpropertiescomparedwithpreviouslydescribedEVswithantibacterialeffect(aEVs,generatedonspecificactivation)orproducedspontaneously(sEVs).The3vesiclepopulationsoverlappedinsizeandinpartoftheconstituentproteinswerestainedwithannexinVandwereimpermeabletoPI.However,noneofthemproducedsuperoxide.Incontrast,remarkabledifferenceswereobservedinthemorphology,abundanceofproteins,andantibacterialfunction.EVsformedspontaneouslyin30min(sEVs)weremoresimilartoEVsreleasedduringspontaneousdeathin1–3dthantoEVsformedin30minonstimulationofopsoninreceptors(aEVs).SpontaneouslygeneratedEVshadnoantibacterialeffectdespitetheirlargenumberandproteincontent.Wehypothesized2parallelmechanisms:onethatproceedsspontaneouslyandproducesEVswithoutantibacterialeffectandanotherprocessthatistriggeredbyopsoninreceptorsandresultsindifferentialsortingofproteinsintoEVswithantibacterialcapacity.Ourresultscallattentiontothefunctionalandmorphologicheterogeneitywithinthemicrovesicle/ectosomefractionofEVs...
