【 摘 要 】
MΦsareimportantsensorycellsoftheinnateimmunesystemandregulateimmuneresponsesthroughreleasingdifferentcombinationsofcytokines.Inthisstudy,weexaminedwhethercytokinesreleasedbyMΦsinresponsetotheprobioticbacterialstrainGR‐1modulatetheresponsesofDCs.ThecytokineprofilereleasedbyGR‐1‐treatedMΦswascharacterizedbylowlevelsofTNF‐α,GM‐CSF,IL‐6,andIL‐12butveryhighlevelsofG‐CSF.GR‐1CMdidnotinduceexpressionofthesharedp40subunitofIL‐12andIL‐23andcostimulatorymoleculesCD80orCD86orincreaseTcellstimulatorycapacityinDCs.However,inG‐CSFR‐deficientDCsorafterantibody‐mediatedneutralizationofG‐CSF,GR‐1CMinducedIL‐12/23p40productionsignificantly,indicatingthatG‐CSFwithintheGR‐1CMinhibitsIL‐12/23p40productioninducedbyotherCMcomponents.GR‐1CMandrG‐CSFalsoinhibitedLPS‐inducedIL‐12productionatthemRNAandproteinlevels.TheinhibitionofIL‐12productionbyG‐CSFwasatleastinpartmediatedthroughinhibitionofJNKactivation.Finally,splenicDCsofGR‐1‐injectedmiceproducedlessIL‐12/23p40thanthoseofPBS‐injectedmiceinresponsetoLPSexvivo,andthiswasatleastpartiallydependentonexposuretoGR‐1‐inducedG‐CSFinvivo.Altogether,theseresultssuggestthatG‐CSFmodulatestheIL‐12/23p40responseofDCsinthecontextoftheprobioticGR‐1throughMΦ–DCcrosstalk...
