Adoptiveimmunotherapycaninducedramatictumorregressionsinpatientswithmelanomaorviral‐inducedmalignancies,butextendingthisapproachtomanycommoncancershasbeenhamperedbyalackofnaturallyoccurringtumor‐specificTcells.Inthisreview,wedescriberecentadvancesinthegeneticmodificationofTcellsusinggenesencodingcell‐surfacereceptorsspecificfortumor‐associatedantigen.Usinggeneticmodification,themanyfunctionalpropertiesofTcells,includingcytokinesecretionandcytolyticcapacity,areredirectedfromtheirendogenousspecificitytowardtheeliminationoftumorcells.Advancesingenedesign,vectors,andcellproductionarediscussed,anddetailsoftheprogressinclinicalapplicationofthisapproachareprovided...
