【 摘 要 】
IL‐7R,FLT3,andCD43aresurfaceantigensexpressedduringthetransitionfrompro‐Btopre‐BcellsinBM.Tounderstandinteractionsbetweentheirsignalingpathways,weanalyzedspontaneousmouseB‐LBLswithdualMLVintegrationintoStat5aandFiz1orStat5aandHipk2.MLVintegrationresultedinup‐regulationofthesegenesinlymphomacellscomparedwithnormalpro‐BcellsfromtheBM.InlymphomaswithbothintegrationsintoStat5aandFiz1,increasesinphosphorylatedSTAT5Aandexpressionofc‐Myc,atargetgeneofSTAT5A,wereobservedfollowingstimulationoftheFLT3.CloneswiththedualintegrationsgrewfasterinIL‐7andFLT3L‐supplementedmediumthancloneswithStat5aintegrationalone.Ontheotherhand,inlymphomaswithintegrationsintoStat5aandHipk2,increasesinphosphorylatedSTAT5Aandexpressionofc‐Mycwereobservedfollowingcross‐linkingofCD43.Inconclusion,FLT3andCD43signalingpathwaysinvolveSTAT5AviaFiz1andHipk2inB‐LBLs.IdentificationofthedualMLVintegrationsitesinB‐LBLs,therefore,willprovideanexcellenttoolforidentificationofthesignalingpathwaysinB‐LBLs...【 授权许可】
