【 摘 要 】
Argininedepletionviamyeloidcellarginaseiscriticallyinvolvedinsuppressionoftheadaptiveimmunesystemduringcancerorchronicinflammation.Ontheotherhand,argininedepletionisbeingdevelopedasanovelanti‐tumormetabolicstrategytodeprivearginine‐auxotrophiccancercellsofthisaminoacid.Inhumanimmunecells,arginaseismainlyexpressedconstitutivelyinPMNs.WethereforepurifiedhumanprimaryPMNsfromhealthydonorsandanalyzedPMNfunctionasthemaininnateeffectorcellandarginaseproducerinthecontextofargininedeficiency.WedemonstratethathumanPMNviability,activation‐inducedIL‐8synthesis,chemotaxis,phagocytosis,generationofROS,andfungicidalactivityarenotimpairedbytheabsenceofarginineinvitro.Also,profoundpharmacologicalargininedepletioninvivoviaADI‐PEG20didnotinhibitPMNfunctionsinamousemodelofpulmonaryinvasiveaspergillosis;PMNinvasionintothelung,activation,andsuccessfulPMN‐dependentclearanceofAspergillusfumigatusandsurvivalofmicewerenotimpaired.Thesenovelfindingsaddtoabetterunderstandingofimmunityduringinflammation‐associatedargininedepletionandarealsoimportantforthedevelopmentoftherapeuticargininedepletionasanti‐metabolictumortherapy...
