Insightintomolecularandcellularmechanismsofinnateimmunityiscriticaltounderstandviralpathogenesisandimmunopathologyandmightbeexploitedfortherapy.WhereasthemolecularmechanismsoftheIFNdefensearewellestablished,cellularmechanismsofantiviralimmunityareonlyemerging,andtheirpharmacologicaltriggeringremainsunknown.COAMisapolysaccharidederivativewithantiviralactivitybutwithoutcomprehensionaboutitsmechanismofaction.TheCOAMmixturewasfractionated,andprophylactictreatmentofmicewithCOAMpolymersofhighMWresultedinaconversionfrom100%lethalmengovirusinfectiontoanoverallsurvivalrateof93%withoutobviousclinicalsequelae.DifferentialandquantitativeanalysisofperitonealleukocytesdemonstratedthatCOAMinducedaprofoundinfluxofneutrophils.Selectivecelldepletionexperimentspointedtowardneutrophilsandmacrophagesaskeyeffectorcellsintherescueofmicefromlethalmengovirus.COAMwasabletoinducemRNAandproteinexpressionofthemouseneutrophilchemokineGCP‐2.BindingofGCP‐2toCOAMwasdemonstratedinsolutionandconfirmedbySPRtechnology.AlthoughCOAMwasnotchemotacticforneutrophils,COAM‐anchoredmuGCP‐2retainedchemotacticactivityforhumanandmouseneutrophils.Inconclusion,thisstudyestablishedthatCOAMrescuedmicefromacuteandlethalmengovirusinfectionbyrecruitingantiviralleukocytestothesiteofinfection,asproposedthroughtheinduction,binding,andconcentrationofendogenouschemokines.Thesefindingsreinforcetheroleofneutrophilsandmacrophagesascriticalcellsthatcanbemanipulatedtowardantiviraldefense...
