【 摘 要 】
Weinvestigatedhowapoptosispathwaysmediatedbydeathreceptorsandcaspase‐8affectcytokineresponsesandimmunitytoLeishmaniamajorparasites.SplenicCD4Tcellsundergoactivation‐inducedapoptosis,andblockadeofFasL‐FasinteractionincreasedIFN‐γandIL‐4cytokineresponsestoL.majorantigens.Toblockdeathreceptor‐induceddeath,weusedmiceexpressingaTcell‐restrictedtransgeneforvFLIP.Inhibitionofcaspase‐8activationinvFLIPmiceenhancedTh1andTh2cytokineresponsestoL.majorinfection,evenintheTh1‐proneB6background.WealsoobservedincreasedNOproductionbysplenocytesfromvFLIPmiceuponTcellactivation.DespiteanexacerbatedTh2response,vFLIPmicecontrolledbetterL.majorinfection,withreducedlesionsandlowerparasiteloadscomparedwithWTmice.Moreover,injectionofanti‐IL‐4mAbininfectedvFLIPmicedisruptedcontrolofparasiteinfection.Therefore,blockadeofcaspase‐8activityinTcellsimprovesimmunitytoL.majorinfectionbypromotingincreasedTh1andTh2responses...
