【 摘 要 】

The crude extracts of flavonoids were obtained from longan leaves via ultrasonic and purified with macroporous resin of AB-8. Taking rutin as the standard, the amount of flavonoids in crude extracts was measured by ultraviolet spectrophotometer. The concentration of flavonoids after extraction and purification was 0.775 μg/mL. Total flavonoids in the extractum accounted for 77.5%. Total flavonoids accounted for 77.49% of the dry weight of the longan leaves. MTT assay was applied to test the A549 cell activities treated by a series of concentrations of flavonoids (50, 100, 200, 400 and 800 μg/mL) for 24h, 48h and 72h, respectively. The morphological changes of A549 cells treated by 100, 200 and 400μg/mL flavonoids for 48h were observed by inverted microscope and fluorescence microscope, respectively. The results showed that the total flavonoids of longan leaves obviously inhibit the proliferation of A549 cells, with cell shrinkage, nuclear fragmentation and apoptosis occurred.
