Chlamydia psittaci (C. psittaci) is an obligate intracellular, gram-negative bacterium, and mainly causes systemic disease in psittacine birds, domestic poultry, and wild fowl. The pathogen is threating to human beings due to closely contacted to employees in poultry industry. The polymorphic membrane proteins (Pmps) enriched in C. psittaci includes six subtypes (A, B/C, D, E/F, G/I and H). Compared to that of the 1 pmpG gene in Chlamydia trachomatis (C. trachomatis), the diverse pmpG gene-coding proteins of C. psittaci remain elusive. In the present study, polymorphic membrane protein 17G (Pmp17G) of C. psittaci mediated adhesion to different host cells. More importantly, expression of Pmp17G in C. trachomatis upregulated infections to host cells. Afterwards, crosstalk between Pmp17G and EGFR was screened and identified by MALDI-MS and Co-IP. Subsequently, EGFR overexpression in CHO-K1 cells and EGFR knockout in HeLa 229 cells were assessed to determine whether Pmp17G directly correlated with EGFR during Chlamydial adhesion. Finally, the EGFR phosphorylation was recognized by Grb2, triggering chlamydial invasion. Based on above evidence, Pmp17G possesses adhesive property that serves as an adhesin and activate intracellular bacterial internalization by recognizing EGFR during C. psittaci infection

    Purpose Exercise training is crucial to the early intervention of pediatric primary hypertension (PHT). However, much less is known about exercise capacity in this disease. This work investigated the exercise capacity in pediatric PHT and analyzed the factors affecting exercise capacity. Methods The study enrolled children with PHT at the Children's Hospital Capital Institute of Pediatrics between July 2017 and July 2020. The Bruce protocol of the treadmill exercise test (TET) was used to assess exercise capacity. Multivariate ordinal logistic regression and generalized linear models were used to analyze factors affecting exercise capacity. Results Of 190 patients, 146 (76.8%) were male, and the median age was 13 (11, 14). Most children accomplished TET and achieved the submaximal heart rates (189 [99.5%]). Children with lower resting diastolic blood pressure (DBP) and 24 h average diastolic blood pressure (ADBP) could achieve a TET stage of 6 or more, whereas children with higher DBP and ADBP could only achieve a TET stage of 3 ( P all < 0.05). Children with lower DBP and 24 h ADBP were also associated with greater metabolic equivalents (METs; r = −0.237, r = −0.179, P all < 0.05). The completion of TET stages was negatively associated with female (OR = 0.163), younger age (OR = 1.198), greater body mass index (BMI, OR = 0.921), and higher 24 h ADBP (OR = 0.952, P all < 0.05). In addition, METs were negatively associated with female (β = −1.909), younger age (β = 0.282), greater BMI (β = −0.134), and higher 24 h ADBP (β = −0.063, P all < 0.05). Conclusions Exercise capacity was impaired among pediatric PHT patients. Female gender, younger age, greater BMI, and higher 24 h ADBP are independently associated with the exercise capacity in pediatric PHT. These findings may help developing scientific exercise prescriptions for pediatric PHT.

      Increasing numbers of studies have revealed theimmunomodulating effects of radiotherapy whencombined with immune checkpoint inhibitors, as radioimmunotherapy has proven to be a promising treatment [1].Radioimmunotherapy has shown significantly improvedtumor responses than radiotherapy or immunotherapyalone in various malignant tumors [2–4]. It has also beenapplied to cervical cancer in multiple ongoing clinicaltrials (NCT03612791 [5] and NCT02635360 [6]). However,tumor recurrence and metastasis are often unavoidable. Assuch, investigations into radioimmunotherapy-inducedtumor ecosystem evolution are essential for guidingimprovements in treatment strategies that achieve better long-term disease control. To date, several studieshave investigated radiochemotherapy-induced tumorecosystem evolution using bulk RNA-sequencing andimmune staining [7, 8]. However, these findings werelimited owing to the cellular heterogeneity in cancers.Single-cell RNA-sequencing (scRNA-seq) enables thecharacterization of cell compositions and transcriptomicstates in the tumor at single-cell resolution.
