Signaling includes recruitment of nuclear factor-κB activator 1 (ACT1) via SEFIR domains (red ovals). Activation of ACT 1 encoded by TRAF3IP2 allows binding to TRAF6 via TNF receptor-associated factor (TRAF)-binding domains (yellow ovals); this activates NF-κB as well as proinflammatory cytokines.
