Starting with the advent of biosensors in the late 1950s and early 1960s, biosensors have come to play a major role in various aspects of human life, including our environment, food and drink, safety, and health. In particular, the detection of clinically relevant biomarkers for the screening, diagnosing, and monitoring of diseases such as cancer is an important goal in improving the treatment outcome and the quality of life of patients. In this thesis, an overview of biosensors and their applications is followed by a detailed description of how a photonic crystal biosensor platform with the capacity for fluorescence enhancement was combined with an automated multiplexed microfluidic flow system and an automated spot-finding and data generation algorithm for the detection of a cancer biomarker in human serum and plasma.
