【 摘 要 】

Measles is a highly infectious viral disease that typically occurs in childhood. Upon infection, it results in a high fever, runny nose, red and watery eyes and a characteristic rash that covers the entire body and lasts up to a week. Despite having a safe and cost-effective vaccine available for 50 years, measles is still one of the leading causes of death among young children throughout the world, with 226,722 reported cases of measles worldwide in 2012 and 114,900 measles-related deaths in 2014. A majority of these deaths are a result of secondary infections, like pneumonia and diarrhea that occur due to measles-induced immune suppression. Recent studies have found that while infectious virus is cleared within 2-3 weeks of primary infection, viral RNA has prolonged persistence in PBMCs, urine, respiratory secretions and lymph nodes. Lymph nodes contain major immune cells that help mount an effective immune response; they are also the point in the infection cycle from where the virus disseminates systemically. In this study, viral RNA persistence in the lymph nodes is extensively investigated using fluorescence microscopy. This research aims to optimize fluorescence in situ hybridization (FISH) to develop a method to visualize measles virus RNA. The study also optimized a protocol to combine FISH with immunofluorescence (IF) to determine the exact cellular location of viral RNA to obtain a more comprehensive and complete understanding of the sites of viral persistence.
