Theyeast Saccharomycescerevisiae isakeymodelsystemfor studyingofamultitudeofcellularprocessesbecauseofitsamenabilityto genetics,molecularbiologyandbiochemicalprocedures.Ultrastructuralexaminations of this organism, though, are traditionally difficult because of the presenceofathickcellwallandthehighdensity ofcytoplasmicproteins.A seriesofrecentmethodologicalandtechnicaldevelopments,however,has revivedinterestinmorphologicalanalysesofyeast (e.g.[1-3]).Herewepresentareviewofestablishedandnewmethods,from samplepreparationto imaging,fortheultrastructuralanalysisof S.cerevisiae.Weincludeinformation for the use of different fixation methods, embedding procedures, approachesforcontrastenhancement,andsamplevisualizationtechniques, withreferencestosuccessfulexamples.Thegoalof thisreviewistoguide researchers that want to investigate a particular process at the ultrastructural levelin yeast by aiding in the selection of the most appropriate approach to visualize a specific structure or subcellular compartment.
