Pairs (18) of microsatelite primers were usedin this study to detect the genetic relationshipwithin Black Slavonian Pig and between TuropoljePig, Mangalitsa breed and Croatian Wild Pigs. Thesecond goal of this study was to determine phylogeneticrelationships among these breeds and someAsian and European pigs using the mtDNA D-loopsequence polymorphism. The third goal was todetermine the MC1R genotype of Black Slavonianpigs and to find an efficient and simple PCR-RFLPmethod, based on differences in MC1R genotype,to distinguish between purebred Black Slavonianpigs and their crossings with commercial pig breedsand Wild Boars. Aiming to conduct microsatelliteanalysis each animal was genotyped for 18 microsatelitemarkers, chosen based on their quality, size,polymorphism and location on the porcine genomeas proposed by the FAO. Two pairs of primers amplifieda 511-bp fragment of control region betweensites 15 390 and 15 900 (Mit1.F and Mit1.R) and a810-bp fragment between sites 15 825 and 16 634(Mit2.F and Mi2.R) were genotyped for mtDNA.Two primer pairs were used to amplify the majorityof the single exon of MC1R gene aiming to determinateMC1R genotype of Black Slavonian pig. Thefirst pair of primers, MERL1 and EPIG2, was usedto amplify a 428-bp product from the 5’ half of theexon, whereas EPIG1 and EPIG3 amplified a 405-bpproduct from the 3’ half. Our results showed thatthe 18 microsatellites used in this study were useful markers to study genetic diversity among Croatianautochthonous pig breeds. This set of microsatellitesmay be used for identifying individuals and for geneticdiversity studies for selection and conservation of theBlack Slavonian pig, Turopolje pig and Mangalitsabreed. Genetic distances between populations madewith Principal Component Analysis (PCA) methodnoticed that studied populations are mostly clearlygeneticaly defined. mtDNA analysis suggested thatBlack Slavonian and Turopolje pig showed geneticdistances from the other European and Chinese pigbreeds. Black Slavonian and Turopolje pig clusteredtogether. Mangalitza was used in the development ofBlack Slavonian pig and this can explain their presencein clustering. Also, with this work, we wereable to identify a simple PCR-RFLP method, basedon different coat color MC1R gene genotypes, bywhich it is possible to detect potential crossings ofautochthonous Black Slavonian pig with commercialpig breeds and including wild boars.
