【 摘 要 】
Context:Vaccinationisconsideredasthemostcosteffectivemethodforthepreventionofhumandiseases.Forthispreventionmethod,weneedcertainsubstancestoincreaseorboosttheantibodyaswellascell-mediatedimmuneresponseagainstvariousbacterialaswellasviralpathogens.Untilnow,alumwasconsideredasthesafestadjuvantforhumanuse,licensedbytheUnitedStatesFoodandDrugAdministration.Duetothepooradjuvanticityofalum,conventionalvaccinesrequiremultiplerecallinjections,atdifferenttimeintervals,toattainorsustaintheoptimalimmuneresponse.Thepresentreviewdiscussesaboutthenecessityofadjuvantsforvaccines.EvidenceAcquisition:Anumberoffactorssuchasslowreleaseofantigen(depoteffect),moreefficientdeliveryofantigentodraininglymphnodes,non-specificactivationofantigen-presentingcellsorofBand/orT-lymphocytes,increaseduptakeofantigenbyantigen-presentingcellsorincreasedrecruitmentofimmunecellstothesitewheretheantigenispresent,cancontributetoincreasedimmuneresponsestoimmunization.Manyofthesefactorsinvolvetheinteractionofvariousimmunesystemcomponentsandspecificanatomicalfeatures,makingthemdifficulttoreplicateinmodelsystemsinvitro.Results:Despitethedevelopmentofmanypotentadjuvantformulationswithvaccineantigenduringthelast80-90years,aluminumcompoundsarestilltheonlyapprovedadjuvantsusedforroutinehumanvaccines.Conclusions:Basedonpre-clinicalandpreliminaryclinicalobservations,itappearsthattherangeofadjuvantsacceptedforhumanvaccineswillexpandinthecomingyears.
