【 摘 要 】
Humanleukocyteantigen-G(HLA-G)isanonclassicalHLA-classIantigenlocatedonchromosome6.HLA-Gishighlyexpressedoncytotrophoblastcellsatthefetomaternalinterfaceandinvolvedinthedevelopmentofpregnantuterusasanimmuneprivilegedsite.ExpressionofHLA-Gisthoughttohaveacriticalroleintheprotectionofthesemiallogenicfetusfrommaternalimmuneattackduringpregnancy.HLA-GmoleculesbindinhibitoryreceptorsonmaternalTcellsandNKcellsandsubsequentlyinhibittheircytolyticactivities.BecauseofmRNAalternativesplicingofHLA-Gprimarytranscript,theHLA-Gproteinexistsinbothmembrane-bound(HLA-G1toG4)andsoluble(HLA-G5toG7)isoforms.HLA-Ggenecontains15alleles,includingtheHLA-G*0105Nnullallele.Asinglebase-pairdeletionofacytosine(1597delC)resultsinopenreadingframemutation,whichleadstoaprematurestopcodon.TheHLA-G*0105NalleleisunabletogeneratetheHLA-G1,HLA-G5,andHLA-G4isoforms.However,itisstillabletoproduceotherHLA-Gproteins,inwhichexon3isremovedbyalternativesplicing,includingHLA-G2,G3,G6andG7isoforms.HLA-G*0105Nnullallelehasbeendescribedinhealthyadultswithsuccessfulandnormalpregnancies,whichsuggeststhatHLA-GfunctionisnotrestrictedtotheHLA-G1isoform.DescriptionofhealthyindividualhomozygousforHLA-G*0105NallelerecommendsthattruncatedHLA-G2andG3isoformsencodedbynullalleleareabletocompensateforthelackoftheHLA-G1,G4andG5isoforms.ResultsofthenumerousstudiesonthenullalleleofHLA-GgeneindicatedthatitsselectionmayhaveincreasedthefrequencyoftheHLA-G*0105N.StudiesonthenullalleleofHLA-GgenecouldbeusefulindeterminingthefrequencyofgeneticvariantsofHLA-Gallelesindifferentethnicgroups.
