【 摘 要 】

Somaticembryogenesisstudieswerecarriedouton11Turkishlentilcultivars.LentilseedsweresterilizedandgerminatedinPlantGrowthRegulator(PGR)freeMS(MurashigeandSkoog)medium.Cotyledon,hypocotyl,root,shoottip,leafandnodexcisedfromsterilegrownseedlingsandembryoswereexcisedfromsterilizedseedandincubatedinMS,B5(Gamborg'smedium),SH(ShenkandHildebrandt)andWH(White)mediawithdifferentPGRs.Sixof11cultivargavepotentiallyembryogeniccallus.Solidmediastudieswerecarriedonwiththesixcultivars.Fourembryologicallybestcallusproducingcultivarsof6wereusedinsuspensionculturestudies.2,4-D(2,4-dichlorophenoxyaceticacid),NAA(1-Naphthaleneaceticacid),BAP(6-Benzylaminopurine)wereusedindifferentconcentrationsandcombinationstoproducecallusandsomaticembriyo.Zygoticembryowasfoundtobebestpotentiallyembryogeniccallusproducingexplant.MSwithNAAand2,4-Dweredeterminedtobeembryogeniccallusreporoductiveconditions.BAPproducedadventitiousshootratherthanembryogeniccallus.Asaresult,calliweretranferredtoliquidcultureandsomaticembryosofYerliKırmızı,Sazak91,KafkasandPull11cultivarsgavebestglobular,heartandtorpedoshapesembriyosinMSmediumsupplementedwith0.3-1mg/lNAA.
