In vitro Variability of Morphogenetic Responses of Tomato (Lycopersicon esculentum Mill.) Explants under Salt Stress and Thidiazuron as Plant Growth Regulator

Forthepresentinvestigation,sixgenotypesoftomato(LycopersiconesculentumMill.)widelygrowninAlgeriawereselectedandusedasresearchmaterialtostudythemorphogeneticvariabilityunderstressconditionsusingdifferentmediatreatmentsandtwotypesofexplants(cotyledonsandhypocotyls)inordertoevaluatetheirresponsestocallusinduction,embryosproductionandcaullogeniccapacity.Invitrogrowthseedlingsagedfor21daysoldwereusedassourceofexplants.TheseexplantswerecultivatedintoaMS-basedmediumcontainingaconstantconcentrationofTDZ(3mg/L)usedasCytokininsPGR,combinedornotwithfourincreasinglevelsofNaClat0,25,50and75mMforeightweeks.ThisCytokininspredisposestheexplanttostress.Statisticalanalysisindicatedahighlysignificantvariationsinresponsesaccordingthenatureofgenotype,treatmentmedium,explantoriginandinteractionsthereofpossessampleinfluenceonmorphogeneticcharactersundertheseconditions.ResultsshowedthatthemorphogenesisresponsesseemstobedependentNaCllevelsaddedinthemedia,amoderateconcentrationsincluding25and50mMinducescomparedtocontrolmedium.Whereas,mediumMT3hadshownarelativetopoorresponseforthemostgenotypestested.ItisinterestingthatthemoderatecombinationNaCl/TDZwasbetterfortheembryosproduction,whichcanbeuseddirectlyorconvertedintoartificialseedsforpropagationofhighvaluehybrid.Allmosteverytestedexplantweredifferentiatedintocallusbutthefavoriteexplantforcarriedaresearchonthevariabilityunderstressconditionsandembryogeniccapacityisthecotyledon.Differencesamonggenotypesweredetected.Foralleffectsanalyzed,homogeneousgroupswereestablishedusingNewman-Keulspost-hottest.ThisattemptmaybeusedasefficientprotocolforgeneticimprovementandtheuseofcombinationPGRs/Stressoragentcouldbetheoptionforthesomaticembryogenesisinvestigations.
