【 摘 要 】
Lipasesholdsignificantpositioninthefieldofbiotechnologyduetoitsabilitytocatalyzeesterification,interesterification,andtransesterificationreactioninnonaqueousmedia.Inthepresentstudyfourdifferentlipaseproducingbacteriawereisolatedfromeffluentsofleather,marbleandtextileindustry.Allthestrains(L9,L15,L16andL20)isolatedweregramnegativeandsubjectedtobothbiochemicalandmolecularcharacterization.BLASTresultsfor16SrDNAsequencesrevealedthatL9(KJ748674)strainhad99%similaritywithAcinetobacterlwoffi.L15(KJ748675)andL16(KJ864927)strainsshowed98%and99%sequencehomologywithAcinetobacterjunniandstrainL20(KJ873872)belongstoAlishewanellaagrisp.Forenzymeproductionstudies,pHandtemperaturewereoptimizedontwodifferentenrichmentmedia.Comparisonoftwodifferentenrichmentmediashowedthatmediumcontainingpeptone,yeastextract,NaClandoliveoilgaveoptimumproductionforallthestrains.AlishewanellaagriL20(isolatedfromtextileindustryeffluent)hadgrowthatalltemperatures(30,37,42and45°C)inmediumcontainingyeastextract,NaClandoliveoil.Incomparisontoallthereportedstrainsofthepresentstudy,Alishewanellaagri(L20)hadmaximumenzymeproductionof17U/mlatelevatedtemperatureof45°C.PartialpurificationandSDS-PAGEanalysisshowedthatproteinbandsoflipaseswererangedbetween35-66.2kDa.Itisevidentthathighcostoflipasehasgenerallyreduceditsexploitationatindustriallevel.Therefore,screeningandidentificationoftheselipolyticstrainswillactasasignificantadditiontothedatabaseonlipaseresearchanditsapplicationatindustriallevel.
