【 摘 要 】

Withthepresentedstudy,itisaimedtoexaminetheeffectofdipeptidylpeptidase(DPP)4inhibitorsitagliptin,whichisanoralanti-diabeticmedicine,ontotalantioxidantandoxidantstatusintheratswhichwereinducedwiththeexperimentaltype2diabetes(T2DM)model.Thestudywasplannedtocover24weeks.Inratsthatareleftoutofthecontrolgroup,after8weeksofHigh-FatDiet(HFD)feeding,theexperimentalT2DMmodelwasconstructedbyalowdoubledosestreptozotocin(STZ)injectionperweek.Sitagliptinwasappliedtotreatmentgroupsatadoseof10mg/kg-day.Attheendofthestudy,FastingPlasmaGlucose(FPG),ALT,AST,LDH,HbA1c,insulin,TotalAntioxidantStatus(TAS),TotalOxidantStatus(TOS)levelsandOxidativeStressIndex(OSI),HOMA-IRandHOMA-βindicesweremeasured.ItwasdeterminedthatsitagliptindecreasedFPGandHbA1clevels,increasedinsulinlevels,anddidnotaffectALT,AST,LDH,TOSlevelsandOSIinthediabetictreatmentgroupcomparedtothediabeticgroup.However,itwasidentifiedthatsitagliptindecreasedTASlevelsinpancreatictissueindiabeticrats,anddecreasedTOSlevelsandOSIintheHFDgroup.Itwasdetectedthatindiabeticratssitagliptinbalancesglucosehomeostasisandprotectsbetacells;whereasitincreasesoxidativestressinpancreatictissueandconverselyitdecreasesoxidativestressinpre-diabeticobeserats(HFDgroups).
