【 摘 要 】
Waterlettuce(Pistiastratiotes)isanimportantmedicinalaquaticplantthatisalsousedforphytoremediationorasbioindicator.Itisverydifficulttomultiplytheplantusingmicropropagationtechniquesduetohighcontaminations.Thestudyaimedtoalleviatethisproblemforefficientmultiplicationoftheplantsubsequently.Thestudymadeuseof60%commercialbleach(5%NaOCl)or60%H2O2forsurfacesterilizationfor10minfollowedbyadding500mg/lDuocid®(awidespectrumantibiotic)intheculturemedium.Theresultsshowedthatcommercialbleachwasnotpropersterliserasallexplantsbleached;whereas,60%H2O2basedsurfacesterilizationwasmostoptimumforthepurpose.TheSurfacesterilizedplantswereinvitroculturedonagarsolidifiedorliquidMSmediumcontaining0.05-0.40mg/lkinetin.Centpercentshootregenerationfrequencywasonbothagarsolidifiedorliquidmedium.However,allexplantsonagarsolidifiedorliquidmediumhadtenancytodieturningallofthemtoalbino.Therefore,multiplicationunderexvitroconditionsonliquidMSmediumcontaining0.05-0.40mg/lkinetinorBAPusingunsterilizedshootmeristemswithoutaddingsucrose.Subsequently,theexplantsinduced2.0-40plantletsperexplanton2.0-3.0mg/lkinetinorBAPcontainingculturemedium.Theplantsregeneratedonbothculturemediawerecomparablewithplantsgrowingundernaturalconditionsanddidnotshowanysignofstresscausingalbinismornecrosis.
