【 摘 要 】
ThereisaconsiderableinterestintheapplicationofmoleculargeneticstechnologiesintheformofspecificDNAmarkersthatareassociatedwithvariousproductivitytraitstopromotemoreefficientandrelativelyeasyselectionandbreedingoffarmanimalswithanadvantageforinheritabletraitsofproductionandreproduction.Riverbuffaloisaspeciesofgreateconomicpotential,especiallyindevelopingcountries;itisthemainsourceofmeatandmilkinEgypt.Thisstudyaimedtodetectthegeneticpolymorphismofthreegenes;follicle-stimulatinghormonereceptor(FSHR),luteinizinghormonereceptor(LHR)andestrogenreceptor-α(ERα)geneswhichareconsideredcandidategenesforfertilityandreproductionperformances.ThegeneticpolymorphismdetectionofthesegenesinEgyptianbuffaloeswillenableustoidentifyandsubsequentlyselecttheanimalswithhighfertilityperformances-dependingonthesemolecularmarkers-forbreedingprograms.GenomicDNAextractedfrom100healthyfemalebuffaloeswasamplifiedusingprimersthatweredesignedfromthecattleFSHR,LHRandERαgenesequences.TheamplifiedfragmentsofFSHR(306-bp),LHR(303-bp)andERα(248-bp)weredigestedwithAluI,HhaIandBglIrestrictionenzymes,respectively.TheresultsshowedthatalltestedbuffaloesaregenotypedasCCforFSHRandTTforLHR.RegardingtotheERαgene,18%oftheinvestigatedbuffaloesaregenotypesasheterozygousAGandtheremaininganimals(82%)aregenotypedasGG.Tothebestofourknowledge,thisisthefirstpublisheddataonthegeneticcharacterizationandnucleotidesequencesoffertilitygenesinEgyptianbuffalo.ThenucleotidesequencesofEgyptianbuffaloFSHRC,LHRT,ERαGandERαAallelesweresubmittedtonucleotidesequencesdatabaseNCBI/Bankit/GenBankandhavetheaccessionnumbersJX049145,JQ885687,JQ308795andJQ308796,respectively.
