Theinteractionbetweencholesterolandhumanserumalbuminhasbeeninvestigated.ThebasicbindinginteractionwasstudiedbyUVabsorptionandfluorescencespectroscopy.FromspectralanalysischolesterolshowedastrongabilitytoquenchtheintrinsicfluorescenceofHSAthroughastaticquenchingmechanism.Thebindingconstant(k)isestimatedtobeK=0.214×104M-1at293K.FTIRspectroscopywithFourierself-deconvolutiontechniquewasusedtodeterminetheproteinsecondarystructureandcholesterolbindingmechanisms.TheobservedspectralchangesindicateahigherpercentageofH-bondingbetweencholesterolanda-helix(secondarystructuremotifinHSA)comparedtothepercentageofH-bondingbetweencholesterolandb-sheets(secondarystructuremotifinHSA).
