【 摘 要 】

Currently,theprimarythrustofresearchinanimalgeneticsistheidentificationofgenes(so-calledmajorgenes),whichaffecttheexpressionofquantitativetraitsmarkedly.Twoofthesemajorgenesaregrowthhormone(GH)anditsreceptor(GHR)geneswhicharerelatedtoproductionandreproductiontraitsinlivestock.ThereisextensiveliteratureonthegeneticpolymorphismofGHandGHRincattle,butperusalofliteraturehasindicatedpaucityofinformationonthesetwogenesinbuffalo.ThisstudyaimedtoevaluatethegeneticpolymorphismwithingrowthhormoneanditsreceptorgenesinEgyptianbuffalousingPCR-RFLPtechnique.GenomicDNAextractedfrom100healthybuffaloeswasamplifiedusingprimersthatweredesignedfromthecattleGHandGHRgenesequences.AllbuffaloanimalsinvestigatedinthisstudyaregenotypedasLLforGHgenewherealltestedbuffaloDNAamplifiedfragmentsat211-bpweredigestedwithAluIendonucleaseandgavetwodigestedfragmentsat159-and52-bpduetothepresenceoftheAluIrestrictionsiteatposition52^53(AG^CT).TheamplifiedfragmentsofGHRgeneobtainedfromalltestedbuffaloDNAat342-bpweredigestedwithAluIendonuclease.TheresultshowedthatalltestedbuffaloesaregenotypedasGGforGHRgene,whereamplifiedfragmentsweredigestedintotwodigestedfragmentsat241-and101-bpduetothepresenceofAluIrestrictionsiteatposition101^102(AG^CT).
