【 摘 要 】

Estrogenreceptoralpha(ER-α)belongtothenuclearreceptorsuperfamilyofligand-inducibletranscriptionfactors.Itisinvolveinbothmammarydevelopmentandinbreastcarcinogenesis.Manypolymorphismssuggestedassociationwiththeincidenceofbreastcancer.ThemostwidelystudiedPolymorphismsarePvuIIandXbaIrestrictionsites,theylieswithinintrononeofestrogenreceptoralphagene.Thisstudywasdonetoevaluatetherelationshipoftwowell-studiedpolymorphismsintheestrogenreceptoralphagene,(PvuIIandXbaI),andtheriskofbreastcancerinJordanianwomen.Inourstudy,100bloodsamplesfromJordanianwomenwithbreastcancerhadbeenobtainedandusedforsubsequentDNAextractionandgenotypedforthePvuIIandXbaIpolymorphismsusingRestrictionFragmentLengthPolymorphism(RFLPs)technique.Also,a100bloodsamplesfromnormalJordanianwomenhadbeenusedascontrol.Forstatisticalanalysis,SPSScomputerizedprogramtocalculateP-valueforpolymorphismsandHardy-Weinbergequationforgenotypefrequencieswasused.TherewasnosignificantdifferencebetweenbreastcancerpatientsandhealthycontrolindividualsinthedistributionofthePvuIIgenotypes.TherewasnosignificantdifferencebetweenbreastcancerpatientsandhealthycontrolindividualsinthedistributionofthePvuIIgenotypes(P>0.05;P=0.27),noassociationbetweenXbaIpolymorphismandbreastcancerrisk(P>0.05;P=0.111).Alsotherewasnosignificantassociationwithhaplotypeanalysis(P>0.05;P=0.07)exceptforAGTChaplotype(P≤0.05;P=0.043)andAATChaplotype(P≤0.05;P=0.027).ThisresultsuggeststhatPvuIIandXbaIpolymorphismdoesn’tplayaroleinthesusceptibilitytobreastcancerinJordanianwomen.
