【 摘 要 】
Theanalysisofcellcycleincancercelllinesprovidesvaluableinformationforthediagnosisofcancer.Cellcyclesynchronizationisaneffectivewaytoanalyzedifferentcellcyclephases.Inthisstudy,bothasynchronousandsynchronousER(+)MCF-7andER(-)MDA-MB-453breastcancercellswereanalyzed.CellswerestainedusingPIandasynchronouscellcyclephaseswereanalyzedbyflowcytometer.ThenbothcelltypesweresynchronizedclosetoG1-Sphaseusing2mMHydroxyureaandsynchronouscellcyclephaseswereanalyzed.Theasynchronouscellsmovedsmoothlyandcompletedtheircyclein24h.ThesignificantinhibitionoftheDNAsynthesisinsynchronousMCF-7cellswasobservedat12haftertheremovalofHydroxyurea,andalmost28%ofthecellswereatSphasewhereasonly58%ofthecellswereinG1phase.AmarkedG1/SaccumulationalsowasobservedinsynchronousMDA-MB-453cellsimmediatelyafterremovalofHydroxyureaat0h.TheDNAsynthesiswassignificantlyblockedbecause%82ofthecellswasatG1phasewhereasonly%10ofthecellswasatSphase.DNAsynthesisresumedandcellsreturnedtonormalcellcycleoverthenext48h.Therewasnosignificant“subG1peak”indicatingapoptosisorcelldeath.ThisdataindicatesthatHydroxyureamaybeusedtosynchronizebothcellswithoutinducingapoptosisorcelldeath.
